What is the best amendment for 3 year old horse manure to grow flowers such as lilies and daisies?

Composted horse manure is a good fertilizer to add to soil, but it is not a good medium for growing plants. Add a couple of inches to your garden and dig it in.

Lilies need well drained, fertile soil. Daisies are very unfussy.

I always add a little lime (no, not the fruit!) to help neutralize the pH. Mix it in, water, and let it sit a day or two before planting.
